"Sono al lavoro, adesso" is the correct form.

"Sono sul lavoro" is wrong. It is accepted in some parts of Italy (as well as "Sono a lavoro"), in speech, but it should not be used.

Another common way to put it is 'sono sul lavoro'; it sounds like 'I'm on work' but it means exactly what you asked. Think of it like 'I'm on shift,' if that helps.

Adding to my previous answer, "Ora sono sul lavoro" could also be thought of as "I'm on the job now." But the meaning is exactly that: "I'm at work now."
